Project B03 will prepare tailor-made polyplexes consisting of guanidinium group-bearing polydepsipeptides and DNA / siRNA (nuclear factor kappa B-decoys and -siRNA (RelA)) that can be applied for the treatment of inflammation related diseases. The characteristics of the polymers will be systematically altered to reach the maximum transfection efficacy in inflammatory vascular endothelial lesions, which will be studied using (i) static models, (ii) dynamic fluidic systems and (iii) in vivo models of the chick area vasculosa of hen eggs. A special focus will be placed on the influence of the protein corona and the blood flow on the performance of the polymers.
